CpG islands is the primary code of chromatin opening at mammalian promoters

Abstract :

The rules governing accessibility and chromatin opening of promoters are crucial to understand the complex regulation of gene expression. We show that CpG islands are major promoter elements in mammalian cells, able to govern open chromatin conformation and support paused transcription. Importantly, nucleosome depletion correlate with CGIs both in vivo and in vitro and in a transcription independent manner. We propose a model whereby CGIs are an essential code for accessibility and gene recognition by the transcriptional machinery that could be regulated by repressive nuclear complexes such as Polycomb.
