Contexte scientifique

Comprendre les principes de construction de l’architecture des plantes : des molécules à l’organisme.

La recherche dans l’équipe SIGNAL touche à une question centrale en biologie du développement : comment des interactions non linéaires à des échelles multiples (molécule, cellule, tissu, organisme) produisent-elles des comportement cellulaires coordonnés et l’émergence de formes précises chez les organismes multicellulaires ? Plusieurs de nos projets sont connectés à la phyllotaxie, cette organisation élégante des feuilles, branches ou fleurs autour des tiges qui montre une régularité frappante et des propriétés mathématiques intrigantes (voir cet article pour plus de détails : Pour la Science).

Pour appréhender cette question, nous utilisons des espèces modèles bien connues : une plante à fleurs (Arabidopsis thaliana) et des mousses (notamment Physcomitrella patens). Pour comprendre comment les dynamiques développementales sont conditionnées par le génotype et conditionne le phénotype, nous combinons des approches issues de différentes disciplines : génétique fonctionnelle, phénotypage quantitatif à différentes échelles (par ex. l’imagerie par microscopie confocale), observations en temps réel et modélisation informatique.

Plus spécifiquement, nos thèmes de recherche portent sur :
 Les signaux hormonaux et leur rôles dans les dynamiques développementales dans le méristème apical caulinaire
  Le contrôle génétique et la robustesse de l’architecture de l’inflorescence
  Le contrôle hormonal et l’évolution des formes ramifiées

Signaux hormonaux et dynamiques développementales dans le méristème apical de la tige

Nous analysons comment les distributions spatio-temporelles de signaux hormonaux et l’activité de signalisation hormonale des cellules contribuent à la formation récurrente de motifs développementaux au niveau du méristème apical caulinaire (le tissu contenant les cellules souches à l’origine des nouveaux organes aériens).

Pour cela, nous suivons plusieurs voies de recherche complémentaires visant à :
 Visualiser de manière quantitative la distribution spatio-temporelle de l’auxine pour mieux comprendre la façon dont elle est est contrôlée ;
 Analyser le réseau de gènes qui contrôle la réponse à l’auxine dans le temps et l’espace ;
 Comprendre comment les connexions symplastiques contribuent à la distribution des hormones et la formation des motifs développementaux ;
 Comprendre comment les hormones contrôlent la croissance au niveau du méristème apical de tige et comment cela contribue à la dynamique de mise en place des motifs développementaux ;
 Comprendre les mécanismes moléculaires permettant à l’auxine de déterminer les identités cellulaires ;
 Décrypter la base moléculaire des interactions auxine-cytokinine pendant la mise en place des organes dans le méristème apical caulinaire.

Nous utilisons des approches allant de l’imagerie quantitative, la génomique, la génétique à la biologie synthétique et la modélisation informatique.

Contrôle génétique et robustesse de l’architecture de l’inflorescence

Cet axe de recherche vise à identifier de nouveaux mécanismes génétiques contrôlant l’architecture de la partie aérienne des plantes sans a priori. Pour cela, nous recherchons d’abord des plantes avec des phénotypes altérés provenant de différents fonds génétiques, indiquant soit une perturbation du méristème apical caulinaire et/ou un changement de la robustesse du caractère (la variance du caractère est alors modifiée). Ensuite, nous cherchons à identifier la base génétique expliquant ces phénotypes altérés. Nous espérons soit découvrir de nouveaux gènes, soit jeter un regard neuf sur la façon dont des gènes connus agissent dans la mise en place de l’architecture de la tige. Lorsque cela est possible, nous travaillons avec des variants génétiques naturels. En effet, ceux-ci nous permettent de questionner l’universalité des mécanismes identifiés chez les lignées cultivées en laboratoire. De plus, ils indiquent de manière plus réaliste comment des changements phénotypiques ont effectivement lieu dans des populations naturelles, voire à plus long terme au cours de l’évolution entre espèces.

Nos projets en cours sont :
  Génétique directe de mutants d’Arabidopsis thaliana affectés dans l’organogenèse et la phyllotaxie
Nous étudions actuellement des mutants non caractérisés affectés dans l’organogenèse et la phyllotaxie. Ces mutants ont été isolés dans la collection de Versailles et sont dans un fonds génétique différent de celui habituellement utilisé (Columbia‑0).

  Variation naturelle de la robustesse de la phyllotaxie
Malgré son apparence régulière, nos travaux ont montré que la phyllotaxie de l’inflorescence montre des variations substantielles dans différents fonds génétiques. L’étude de ces variations peut révéler en filigrane les "règles" de formation de la phyllotaxie et indiquer comment les comportements cellulaires sont coordonnés dans l’espace et le temps pour générer des motifs développementaux réguliers. Nous analysons actuellement différentes accessions naturelles pour caractériser les niveaux de perturbations de leur phyllotaxie.

  Coordination entre identités d’organes dans une transition de phase rapide
Les plantes à fleurs passent rapidement d’un état végétatif à floral quand elles commencent à produire des fleurs. Chez les Brassicaceae, comme la plante modèle Arabidopsis thaliana, les tiges arrêtent de produire des unités de base comprenant une feuille et une branche pour faire seulement des fleurs. Aucune forme intermédiaire n’est produite. Un faisceau d’éléments suggère que la feuille (aussi appelée bractée) est inhibée lorsque la fleur se met en place, via des mécanismes inconnus ayant évolués chez les Brassicaceae.
Nous avons identifié un fonds génétique chez Arabidopsis chez qui certaines bractées sont associées à des fleurs lors de la transition florale. Nous caractérisons actuellement les variations génétiques qui perturbent la coordination robuste entre bractées et fleurs au cours de la transition florale.

Contrôle hormonal et évolution des formes ramifiées

Un objectif central en biologie est de comprendre quels changements développementaux et génétiques ont contribué à l’évolution des formes dans le temps. Chez les plantes, les diverses positions des branches, résultant de l’activité coordonnée des méristèmes aux extrémités des pointes de tiges, constituent un facteur déterminant de la diversité des formes. Il a été démontré que des signaux hormonaux jouent un rôle majeur dans le contrôle de la ramification dans différentes lignées de plantes. Ici, nous cherchons à comprendre (1) comment l’interaction entre les signaux hormonaux régule la distribution des branches et (2) la manière dont cette interaction a été remodelée pendant l’évolution afin de générer la diversité architecturale observée chez les plantes. Nous utilisons les mousses comme organismes modèles.

  Transport apolaire de l’auxine chez la mousse Physcomitrella patens
La distribution des branches sur les tiges est coordonnée à l’échelle de la plante entière par trois signaux hormonaux : auxine, cytokinine et strigolactone. Ces molécules existaient avant la divergence évolutive des mousses et des plantes à fleurs, et ont été recrutées indépendamment dans les deux lignées végétales pour contrôler la ramification des tiges feuillées. Chez les plantes vasculaires, comme Arabidopsis thaliana, le transport longue distance de l’auxine est médié par les protéines PINs et est un régulateur clef de la ramification. Chez la mousse Physcomitrella, des expériences utilisant inhibiteurs pharmacologiques et mutants ont montré que les homologues aux gènes PINs ont un rôle mineur dans la ramification des tiges. En revanche, le transport d’auxine apolaire et diffusif a été proposé comme un régulateur de la ramification chez la mousse. Ce mécanisme pourrait être médié par les plasmodesmes, dont l’ouverture est activement régulée par des dépôts de callose. Nous étudions actuellement la base génétique de ce mécanisme chez la mousse.

  Développement et évolution des formes ramifiées chez la mousse
Notre recherche a montré que trois signaux hormonaux régulent la ramification des tiges et ont un rôle conservé chez des espèces évolutivement distantes. Cependant, on ne sait toujours pas comment les mêmes signaux peuvent générer la diversité des formes ramifiées observées dans la nature. Nous cherchons à résoudre ce problème biologique fondamental en utilisant la lignée évolutive des mousses comme modèle, et une approche multidisciplinaire combinant modélisation informatique et génétique du développement.

Solutions d’imagerie multi-échelles pour l’analyse quantitative des formes

Un facteur limitant pour comprendre la dynamique de l’émergence des formes est la capacité à quantifier des paramètres développementaux dans le temps et l’espace à différentes échelles. Pour lever cette limitation, l’équipe SIGNAL a développé et continue à concevoir :
 Des biosenseurs hormonaux ;
 Une solution robotique open-source pour automatiser le phénotypage des parties aériennes des plantes (comme Arabidopsis) avec une haute précision, et un débit raisonnable
 Des solutions informatiques pour l’analyse quantitative d’images
