Parkinson disease (PD) is the most common age-dependent neurodegenerative movement disorder. Accumulated evidence indicates both environmental and genetic factors play important roles in PD pathogenesis. Levodopa can temporarily relieve some PD symptoms, but there is currently no treatment to halt disease progression or prevent neurodegeneration. Our long-term goal is to develop effective treatment by understanding the pathogenic mechanism of the disease. In the last decade or two, we have demonstrated that PD-associated PINK1 and parkin forms a physical complex and that PINK1 is required for activity of parkin E3 ligase. We have further shown that PINK1 phosphorylates Drp1 to regulated mitochondrial fission. This regulation is impaired in cells derived from PD patients harboring PINK1 mutations. We have recently identified alas1 as a novel PD risk factor and demonstrated that alas1 plays important roles in maintaining DA neuron survival and regulating system aging. These findings lay the foundation for study of PD etiology.
