Zymolyase Stocks 

We prefer Zymolyase to Lyticase, but this is totally subjective... 

Zymolyase: ref 120491 from SEIKAGAKU distributed by MPbiomedicals (20U/mg)

The enzyme is unstable once resuspended in liquid, so it is safe to store it as small aliquots of powder. Considering the high price, it is more effective to mix the powder to sorbitol powder (in most cases, the presence of sorbitol does not interfere with the lytic activity and downstream reactions) and aliquot this mixture. This way, very small amounts of enzyme can be aliquoted in each tube.

 

 

Mix thoroughly 50mg of Zymolyase powder with 2275 mg  of sorbitol (molecular biology grade).

Aliquot 46.5 mg of this mixture in individual eppendorf tubes (make sure you tare each time) and store at 4°C.

 

Before use: resuspend in 500 ul water to obtain a solution of 2mg/ml Zymolyase, 0.5M Sorbitol.
