Comment les cellules communiquent-elles entre elles et avec l’environnement?


Les plantes sont fixées au sol. Elles doivent donc adapter constamment leur croissance et leur architecture en fonction des changements de leur environnement. Au sein de l’organisme, les cellules intègrent en permanence ces signaux, les traduisent en décisions cellulaires qui vont déterminer le destin de la cellule et permettre de former de nouveaux organes ou de changer leur forme. Pour cela, les cellules sont équipées de molécules, appelées récepteurs, qui permettent la communication entre cellules.


La membrane plasmique est un compartiment clefs dans la signalisation cellulaire et pour les échanges avec le milieu extérieur. Les phospholipides anioniques (e.g. phosphatidylinositol phosphates ou PIP) sont des lipides minoritaires mais qui ont un rôle crucial pour établir  l’identité de la membrane plasmique (par le recrutement de protéines interagissant spécifiquement avec ces lipides, mais aussi de par leurs propriétés biophysiques particulières). De plus, leur production est très finement régulée, à la fois dans le temps et dans l’espace, ce qui en font des régulateurs clefs de la signalisation cellulaire.


Nous abordons les questions suivantes :


-Quels sont les lipides anioniques qui contrôlent l’organisation de la membrane plasmique et comment?


-Quelles sont les fonctions des lipides anioniques dans la signalisation hormonale ?

(e.g. brassinosteroid et auxin)


-Comment les lipides anioniques régulent-ils l’identité des compartiments endomembranaires et le traffique vésiculaire entre ces compartiments ?


- Comment l’homéostasie des lipides anioniques est-elle dynamiquement régulée par les variations de l’environnement de la plante et contribue ainsi à son adaptation?



 

Notre recherche est basée sur des approches de biologie cellulaire et du développement tel que la microscopie confocale, l’imagerie du vivant, l’analyse de patron d’expression, l’analyse de mutants, la génétique ainsi que la biochimie des protéines
