L’une des barrières principales à l’autofécondation est l’auto-incompatibilité (AI) permettant à l’organe reproducteur femelle, le pistil, de discriminer le pollen « soi » du pollen « non-soi ». Chez les Brassicacées, la reconnaissance du pollen « soi » repose sur une interaction allèle-spécifique entre un récepteur et son ligand. Le gène SRK (S-locus Receptor Kinase) code un récepteur transmembranaire à activité kinase exprimé spécifiquement dans les cellules épidermiques du pistil (papilles stigmatiques). Le gène SCR (S-locus Cysteine Rich) code un peptide sécrété d'environ 80 résidus, localisé dans la paroi du grain de pollen. Lors de l’autopollinisation, le peptide SCR traverse la paroi des papilles stigmatiques pour atteindre le récepteur SRK provoquant alors son autophosphorylation et l’activation de la voie de transduction conduisant au rejet du pollen.
Nous avons récemment montré par immunolocalisation, que le récepteur SRK est majoritairement présent dans des endosomes et que seule une petite fraction est détectée au niveau de la membrane plasmique (Ivanov & Gaude, 2009). Cependant, cette faible proportion de SRK à la membrane semble être suffisante pour fixer le ligand ; le récepteur SRK est alors rapidement internalisé après perception du signal.
Nous voulons désormais aborder les questions suivantes :
-Quel est le rôle du traffic intracellulaire dans la réponse d’AI ?
-Comment une papille stigmatique peut-elle discriminer en même temps auto- et allo-pollen ?
