Notre travail vise à répondre à une question centrale en biologie du développement des plantes : comment les informations positionnelles fournies par les signaux hormonaux, des régulateurs morphogénétiques essentiels chez les plantes, régulent la mise en place des patrons spatio-temporels d’identités cellulaires au cours du développement ?
Pour cela nous utilisons des approches pluridisciplinaires et quantitative afin d’étudier comment la distribution dynamique des hormones végétales contrôle l’expression des gènes dans le méristème apical caulinaire.

PRINCIPAUX RESULTATS

1- Nous avons développé un nouveau senseur de la signalisation auxine, DII-VENUS, qui permet d’estimer la distribution de l’auxine dans des tissus vivants avec une haute résolution spatio-temporelle (Brunoud et al., Nature 2012).
Ce travail a été réalisé en collaboration avec Malcolm Bennett (U. Nottingham), Christian Hardtke (U. Lausanne), Klaus Theres (MPI Cologne), Jose Dinneny (Carnegie Institution at Stanford) et Christa Testerink (U. Amsterdam).

2- Nous avons démontré qu’il existe un contrôle spatial de la signalisation auxine au sein des cellules du méristème d’inflorescence, cellules qui jouent un rôle clé dans la mise en place de la phyllotaxie (Vernoux et al ., MSB 2011).
Ce travail a été réalisé en collaboration avec Lieven de Veylder (VIB Ghent), Christophe Godin et Yann Guédon (INRIA/CIRAD Montpellier)

3- Nous avons contribué à élucider les bases structurales des interactions protéiques clé de la signalisation auxine. Les interactions Aux/IAA-ARF sont contrôlées par un domaine conservé, le domaine III-IV. Nous avons obtenu la structure du domaine III/IV d’ARF5 par cristallographie et démontré que ce domaine est de type PB1, ayant une face globalement positive et une autre négative (Nanao et al., Nature Com 2014).
Ce travail a été réalisé en collaboration avec François Parcy, Renaud Dumas (PCV Grenoble) et Max Nanao (EMBL Grenoble).

4- Nous avons identifié un champ inhibiteur de la signalisation cytokinine nécessaire à la robustesse du plastochron du méristème d’inflorescence. Nous avons découvert une fonction non cellule autonome de la protéine AHP6, un régulateur négatif de la signalisation des cytokinines, dans la mise en place de la phyllotaxie (Besnard et al., Nature 2014).
Ce travail a été réalisé en collaboration avecYkä Helariutta (U. Helsinki), Arezki Boudaoud (ENS Lyon), Christophe Godin et Yann Guédon (INRIA/CIRAD Montpellier).

5- Nous avons généré un senseur pour les jasmonates permettant d’étudier la dynamique de la réponse à la blessure chez les plantes ainsi que leur distribution à l’échelle cellulaire (Larrieu et al., Nature Com. 2015).
Ce travail a été réalisé en collaboration avec Laurent Laplaze (IRD, Dakar & Montpellier), Antony Champion (IRD, Dakar & Montpellier) et Malcolm Bennett (U. Nottingham).
