High efficiency sporulation in liquid

Streak your diploid a/alpha S.cerevisiae strain on a fresh YPD plate. (It is important to start the sporulation from a fresh colony).

Inoculate a YPD culture with a colony, grow 24h at 30°C.

Spin down the cells, resuspend them in YPA medium and culture them for 8 hours at 30°C

Spin down the cells, resuspend them in 2% KAc medium, and incubate at 30°C for 5 days. For some mysterious reason, it is important to set your tube in the incubator with a 45° slope.

YPA medium:

10g/L Bactopeptone

10g/L Bacto Yeast Extract

2%     KAc
