Title : HMGB1 AS AN ARCHITECTURAL CHROMATIN PROTEIN AND EXTRACELLULAR SIGNALLING MOLECULE
High-mobility group box protein 1 (HMGB1) is a non-histonenuclear protein that has a dual function. Inside the cell, HMGB1 acts as an architectural chromatin-binding factor that binds and bends DNA and promotes protein assembly at specific targetsregulating transcription and determining chromosomalarchitecture. Outside the cell, HMGB1 serves as an alarmin toactivate the innate system and mediate a wide range of physiological and pathological responses. HMGB1 can interact with receptors that includeRAGE (receptor for advanced glycation endproducts) as well asToll-like receptor-2 (TLR-2) and TLR-4 and function in a synergisticfashion with other proinflammatory mediators to induce responses.HMGB1and RAGE are considered as a ligand–receptor pair that plays an important role in regulating theinvasiveness of tumor cells. RAGE is downregulated in adult life except for skin and lung. HighRAGE levels were observed in different tumors while decreased amounts were found in lung carcinomas.
