Developmental Control of Replication Timing and Chromosome Architecture
Abstract :
All eukaryotic cells replicate segments of their genomes in a defined temporal sequence. This temporal sequence ("replication timing program") is developmentally regulated, evolutionarily conserved, and is disrupted in many diseases and possibly all cancers. However, both the mechanism and the significance of this program remain unknown. Using several approaches, we have found a strong relationship between replication timing and the 3D organization of chromatin in the nucleus. I will summarize these data and our recent findings suggesting that units of developmental regulation of replication timing reflect a previously unappreciated unit of large-scale chromosome structure and function.
