Meiotic recombination is initiated by programmed DNA double-strand breaks (DSBs), but their number and distribution must be carefully controlled spatially and temporally to ensure successful chromosome segregation while limiting genome instability. In budding yeast, we are using simulations (a) to investigate how the DNA damage signalling kinase Tel1 regulates the patterning of meiotic DSBs, providing negative feedback that shapes where and how frequently Spo11-dependent breaks are formed.
I will discuss this work alongside our studies of the Top3–Sgs1 pathway, which regulates the movement and processing of meiotic recombination intermediates. By directly mapping Top3 catalytic activity, our work (b) reveals how transcription strongly influences the dynamics of recombination intermediates. Together, these studies explore how the spatial organisation of chromosomes and local regulatory mechanisms shape where meiotic recombination begins and how it proceeds.
