The growing success of monoclonal antibodies (mAbs) in therapy highlights the importance of these molecules for human health. To identify candidate mAbs for new targets, a common pipeline involves the selection of antibodies produced by an immunized mouse and the grafting of murine complementarity-determining regions (CDRs) onto human frameworks to reduce immunogenicity, a process called humanization. However, the underexplored effects of this process on antibody dynamics and the challenge of producing stable mAbs raise the question: “What are the structural influences of CDRs beyond their role in antigen binding?”
In this presentation, we will explore through molecular dynamics simulations how CDRs can affect antibody conformation and dynamics. We will also discuss the relationship between these effects and mAb stability and introduce our next steps toward understanding their impact on antibody developability.
