DNA Quantification using

« Qubit ds DNA BR Assay kit »

Using :

Qubit ds DNA BR Assay kit (Invitrogen : ref : Q-32850)

Qubit assay tubes (set of 500) (Invitrogen : ref : Q-32856)

For best results, ensure that all assay reagents are at room temperature before you start.

1- Set up two Assay Tubes for the standards and one tube for each sample.

2- Prepare the Qubit Working solution by diluting the Qubit reagent 1/200 in Qubit buffer. Prepare 200uL of Working solution for each standard and sample.

3- Prepare the Assay Tubes as follow :

For Standards : 190uL of Working solution (from step 2) + 10uL of Standard (from kit)
For Samples : 198uL of Working solution (from step 2) + 2uL of Sample

4- Vortex all tubes for 2-3 seconds.

5- Incubate the tubes for 2 minutes at room temperature.

6- Insert the tubes in the Qubit 2.0 Fluorometer and take readings.

Optional : Using the Dilution Calculator feature of the Qubit 2.0 Fluorometer, determine the stock concentration of your original sample.
