RNA is a dynamic molecule that can fold into many different conformations. Indeed, the biological function of an RNA is often related to this conformational flexibility. A key question in my group is to determine to which extent the strength of RNA base pairing interactions and conformational dynamics of the stimulatory structure defines the propensity to frameshift, move to an alternative reading frame on certain messenger RNAs during translation. Such alternative translation mechanisms are crucial for celular as well as for viral genes. Towards this goal, my group employs fluorescence and force-based single-molecule and ensemble kinetic analysis approaches to uncover fine mechanistic details and RNA and protein dynamics at a high-spatiotemporal resolution. In my talk I will present our recent work on HIV-1 RNA translational dynamics focusing on global and single molecule analysis of HIV-1. Our work highlights the complexity of HIV-1 gene regulation and host-pathogen interactions upon HIV-1 infection. Furthermore, we provide detailed insights into a new RNA structural fold including the frameshift site, which could impact viral replication and could serve as a target for antiviral therapy.
