Multigenerational epigenetic variation : an epigenomics and quantitative genetics perspective
Abstract :
DNA methylation patterns tend to be propagated across generations in plants, rather than re-established anew at each generation like in mammals. As a result, plants appear particularly prone to the inheritance of epigenetic changes. In order to assess the potential impact of such epimutations on heritable phenotypic variation, we have created a population of over 500 epigenetic recombinant inbred lines (epiRILs) in the reference plant Arabidopsis. These lines were derived from an initial cross between two parents with near identical genomes but contrasted DNA methylomes. I will present the results of epigenomic mapping and sequencing of >100 of these lines. This information has provided us with unprecedented insights into the variable degrees of stability of epimutations across generations as well as their impact on complex traits, gene expression, transposable elements mobilization and homologous recombination.
