Transcription-replication conflicts cause R-loop-dependent chromatin alterations

R-loops are three-stranded nucleic acid structures that form co-transcriptionally and which are comprised of an RNA-DNA hybrid and a displaced strand of ssDNA. Although R-loops have important functions in cells, their dysregulation can also lead to genome stability. We recently showed that R-loops forming at engineered transcription-replication conflict sites lead to reduced nucleosome occupancy and replication fork blockage. In this seminar, I will describe our recent studies how these R-loop/TRC-dependent chromatin alterations can also impact the functional state of cells and threaten cellular identity.
