Title : Fluorescence fluctuation microscopy or how to measure concentration and mobility of molecules for cellular biology.

Experimental and conceptual tools of physics have made it possible to raise new questions in biology. This is of course true in many domains and especially in cell biology, thanks to optical microscopy. I will focus on the case of the familly of methods called Fluorescence Fluctuation Microscopy, the goal of which is to provide information about the concentrations and mobilities of molecular species (DNA, proteins and others) implied in various biological phenomena. I will present recent results obtained in our group at LIPhy, thanks to developments in optical engeenering and methodology, among which : *Cellular response to heat shock - monitoring the mobility of the heat shock transcription factor HSF1 ; *adaptive optics : from single molecules and single cells to multicellular spheroids ; *monitoring absolute surface concentrations on substrates ;
